P807 project

PERK is a kinase that plays a fundamental role in the regulation of protein production and degradation, which is particularly important for the survival of tumors that produce and secrete high amounts of proteins, such as antibody-secreting multiple myeloma (MM) in which PERK inhibition induces death of cancer cells due to an effect known as proteotoxicity. NMS-812 is an extremely potent, orally available PERK inhibitor with good efficacy and tolerability in preclinical models of MM, also in combination with standards of care, with potential for first-in-class. Preclinical development of NMS-812 is ongoing, with the aim of starting clinical study of the drug in MM in the near future.
